Rapamycin is a macrocyclic triene antibiotic that binds to and inhibits the molecular target of rapamycin (mTOR). Rapamycin is a potent immunosuppressant used as an alternative to calcineurin inhibitors and has anticancer activity. Sirolimus restricts the proliferation of smooth-muscle cells by blocking cell cycle progression at the G1/S transition.
